HAT medium分子式：CAS号：性质：HAT为次黄嘌呤（hypoxanthine）、氨基蝶呤（aminopterin）和胸腺嘧啶脱氧核苷（thymidine）的缩写。它是根据嘌呤和嘧啶生物合成途径设计的分离杂种细胞的特殊培养基。氨基蝶呤阻断核苷酸的主要合成IMP途径，因此细胞只能通过次黄嘌呤鸟嘌呤核糖磷酸转移酶（HGPRT）使次黄嘌呤磷酸化形成次黄苷酸，依次转化为AMP和GMP，以及通过胸腺嘧啶核苷激酶（TK）使胸腺嘧啶核苷磷酸化形成脱氧胸苷酸，即通过“应急”道路依赖外源的嘌呤和嘧啶来合成核苷酸。用HGPRT—TK+细胞与HGPRT+TK—细胞作亲本进行细胞杂交，在HAT培养基中，未融合的亲本细胞及融合的同核细胞死亡，这是因为这些细胞的核苷酸合成的主要途径被阻断而又不能利用外源核苷酸原料所致，只有融合的细胞由于酶的补偿成为HGPRT+TK+细胞，在HAT培养基中生成而被筛选出来。
