超离心沉降测定质点在重力场中的沉降速度以计算质点大小的分析方法，只适用于粗分散体系。对于质点很小的分散体系（＜0.1微米）,由于质点的扩散作用，重力沉降法已无法应用。但如果把分散体系置于强离心力场中（如用超离心机），则质点的大小仍可用沉降方法来测定。用超离心机测定质点大小时，常用的有沉降平衡和沉降速度两种方法。沉降平衡法 此法采用的离心力场约为重力场的104倍。质点在离心力的作用下向沉降池的底部移动，因而形成浓度梯度。体系中一出现浓度梯度,则发生扩散,而且方向与沉降相反。当扩散与沉降达到平衡后，沉降池中各处的浓度不再随时间而变。这时可根据下式计算单个质点的质量m或高分子的分子量Μ：(1)(2)式中k和R分别为玻耳兹曼常数和气体常数;T为热力学温度；v为质点的偏微比容；ρ为介质的密度；w 是转动角速度；x是沉降池中某一位置离转轴的距离；c1和c2是x1和x2处的浓度。因此,只要在沉降达到平衡后测定沉降池中不同位置的浓度，即可算出质点的大小。沉降速度法 若离心力场强度增至重力场的105～106倍，则扩散作用可以忽略，质点在离心力的作用下运动。当质点所受净的离心力与粘滞阻力达到平衡时，从理论上可导出：(3) (4)式中D为扩散系数；S为沉降系数。S 的物理意义是单位离心力作用下的沉降速度，即：(5)式中dx/dt为在沉降池中x 处质点的沉降速度。若对式(5)进行积分，则得：(6)式中x1和x2为时间t1和t2时质点在沉降池中位置,实际上也是介质与胶体体系的界面在t1和t2时的位置。因此，只要测定不同时间界面移动的位置即可求出 S。但要求出质点的大小则还需要体系扩散的数据。在推导式 (3)和 (4)的过程中，未考虑质点移动时的互相干扰。因此，两式中的D和S均应是无限稀释时的扩散系数和沉降系数。为此，要求出D、S与浓度的关系，然后通过外推方法求出浓度趋近于零时的D和S。
