急性毒作用环境污染物一次或24小时内多次作用于人或动物机体所引起的损害作用。可用动物实验来阐明环境污染物对机体的作用途径、毒性表现和对机体的剂量与效应之间的关系（见剂量－反应关系），并为进行亚急性、慢性以及其他特殊毒性试验的实验设计提供依据。急性毒作用一般以半数有效量 (ED50)来表示，即直接引起一群受试动物的半数产生同一中毒效应所需的毒物剂量。ED50 数值越小,则受试物的毒性越高,反之则毒性越低。半数有效量如以死亡作为中毒效应的观察指标，则称为半数致死量(LD50)或半数致死浓度(LC50)。半数有效量是以数理统计方法计算出预期能引起50％的动物出现同一生物学效应的受试物剂量。它有一定的误差，故常用"可信限"来表示可能的变动范围。急性毒性试验的基本方法，是将一系列不同浓度或剂量的受试物一次或在24小时内多次分别通过口服、灌胃、吸入或经皮肤等途径给予许多组的受试动物。常用的受试动物为成年大鼠或小鼠，一般观察1～4天。如中毒症状出现和恢复迟缓，则可延长到4周。环境污染物毒性根据半数致死量,一般分为5级（见表）。急性毒性试验只能粗略地判定一种受试物的毒性，这往往不够全面。如有些受试物急性毒性较低，但长期接触较小剂量，可因毒作用蓄积等因素而使毒性变高。为了全面评定一种环境污染物，除进行急性毒性试验外，还必须进行亚急性、慢性和其他特殊毒性试验，而急性毒性试验则是其他试验的基础。近年来在进行亚急性和慢性毒性试验之前，往往先进行多次预试验，即以相当于能使动物死亡10％以下的一次剂量为最高剂量，以其若干分之一为低剂量,进行5～21天的试验。这种试验结果往往比急性毒性试验结果更接近亚急性和慢性毒性试验，可为亚急性和慢性毒性试验的剂量分组作出更精确的估计。参考书目WHO, PrinciplesandMethodsfor Evaluating the ToxicityofChemicals, Part 1.Geneva,1978.
