植物抗性基因工程植物抗性基因工程resistance gene engineering of plantszhiwu kangxingjiyln gongeheng植物抗性基因工程(。sistancege脱engineeringof禅ants)根据分子遗传学原理，培育具有特定抗性的植物新品种的生物技术。它的程序是:鉴定和分离抗性基因—抗性基因的重组—将抗性基因导人受体，获得抗性能够表达并稳定遗传的再生个体。植物抗性基因工程，包括植物抗虫基因工程、抗除草剂基因工程和抗逆基因工程。1987年菲斯可夫首次将苏云金杆菌体内的晶体蛋白毒素的基因克隆出来，与花椰菜花叶病毒(CaMy)355启动子构成嵌合基因，以Ti质粒为载体将之转移到番茄植株中并得到表达，鳞翅目昆虫的幼虫取食这种转基因植物后产生厌食等症，几天内死亡。另外筛选的目的基因是抑制蛋白酶的基因，害虫取食转基因植物因消化系统中蛋白酶被钝化，因饥饿而死。植物抗性基因工程包括植物抗病毒、抗真菌等病原生物的基因工程。1986年美国华盛顿大学和孟山都遗传工程公司首次将烟草花叶病毒(TMV)的外壳蛋白基因转移至烟草和番茄中，获得的抗IMV的工程植物已通过大田试验。植物抗除草剂的基因工程是用基因工程方法培育对除草剂不敏感的作物新品种，获得方法一是将某种能以除草剂为底物的酶的基因转人植物，这样的转基因植物能分解除草剂而不致受害。另外是改造植物中除草剂作用的靶物位点或增加靶物产量。以点突变的方法改变靶物中的氨基酸使除草剂不能识别它所要破坏的酶或结构蛋白;将靶物基因转人植物中使之高效合成，以达到抗除草剂的目的。中国科学家利用龙葵中抗除草剂阿特拉津的ISh今基因，育成了抗阿特拉津的转基因大豆和水稻。植物抗逆基因工程是采用基因工程的方法，培育抗盐碱、抗旱、抗涝、抗冻(寒)等不良环境的植物新品种。抗性基因来源于天然的抗逆性植物或微生物中直接分离，也可以是植物在逆境中产生的起保护细胞作用的蛋白质的基因，如热休克蛋白基因。如将这些基因转移到抗逆力弱的品种中，就可以提高抗逆性。(程治英)
